Can I use this to connect 27in iMac to my PC's graphics card and use the iMac as a external monitor?

You should be able to as long as your Video Card has a Mini DisplayPort like the NVIDIA GeForce GT 120 Graphics Upgrade Kit for Mac Pro. This video card will work on a PC. I will be trying this as soon as my 27in iMac arrives...

no, but you can can connect a secondary monitor to your imac from the mini displayport
